Cluster of differentiation 28 (CD28), is a 44 kDa, type I transmembrane (glycoprotein) receptor, predominantly expressed on naïve T cells. Its role is to provide co-stimulatory signals required for T cell activation and survival. Through its counter-receptors; B7 ligands (CD80 and CD86), expressed by antigen presenting cells (APC), CD28 ligation has been shown to enhance the production of various cytokines, such as IL-2, IL-4, and IL-10, and has been implicated in the prevention of T cell anergy. The ligation further stabilizes Bcl-xL, an antiapoptotic molecule required for T cell survival. CD28 was one of the first molecules discovered to play a role in Treg-mediated control of autoimmunity. In essence, CD28-mediated co-stimulation greatly enhances the T cells effector response to antigen.
