The SARS-CoV-2 Nucleocapsid (N) protein is the most abundantly expressed protein of the coronavirus. The N protein is composed of two protein domains connected by an intrinsically disordered region (IDR), known as the linker region. It binds to RNA to form helical ribonucleoprotein (RNP) structures during packaging of the genome into the viral capsid, while also regulating RNA synthesis during replication and transcription. N protein, along with the coronavirus membrane protein (M), facilitates viral assembly, while also modulating host cell metabolism and manipulating the cell cycle. Due to the strong immunogenicity and conserved nature of the coronavirus nucleocapsid, N proteins have value in diagnostic and vaccine development.

Figure 2. Western blot analysis of PtX™ Human Anti-SARS-CoV-2 N (HuN4). Lanes 2 – 5: Varying amounts of SARS-CoV-2 Nucleocapsid (N) Phosphoprotein were run on the SDS-PAGE. Separated bands were transferred to the membrane and PtX™ Human Anti-N (HuN4) (1: 1 000) was used to detect the antigen. The SARS-CoV-2 N Phosphoprotein bands (~46 kDa) were visualized in each lane following the addition of anti-human secondary antibody with HRP and substrate. Our antibody was able to detect antigen amounts upwards of 15.6 ng (Lane 5).
