The structural capsid protein p24, which has a molecular weight of 24 kDa, is the most abundant viral (HIV) protein and forms part of the Gag polypeptide. It is detectable at relatively high levels in the blood during the early stages of HIV infection, making it a potential viral marker for diagnosis, blood donor screening, monitoring disease progression, and evaluation of antiretroviral therapy efficacy. Initial HIV tests only measure antibody levels, which are only detectable approximately 3–4 weeks post-infection. The p24 protein can be detected in patient blood as early as two weeks post infection, making p24 antigen particularly valuable for the diagnosis of primary HIV infection.

Figure 2: Antigen-Antibody binding assay (ELISA). An ELISA plate was coated overnight with 100 ng of HIV-1 recombinant p24 antigen per well. Detection was performed using PtX™ Human HIV-1 p24 Recombinant Antibody (HRP) was applied at a dilution of 1:2 000 and incubated for 60 minutes at 37 °C, following which a colorimetric detection substrate was used for visualization. Absorbance (OD) was read spectrophotometrically at 370 nm. Increasing amounts of HIV-1 p24 antibody were plotted against the corresponding OD units.
