Tubulin is a major component of microtubules, the scaffolding of the eukaryotic cytoskeleton. It plays multiple roles in the maintenance of cell structure, intracellular vesicle and organelle transport, and cell division (mitosis and meiosis). Microtubules are formed by polymerization of alpha and beta tubulin isotypes, and the addition of proteins referred to as microtubule-associated proteins (MAPs). It has a molecular weight of 50 kDa and is composed of a double β-sheet core, enclosed within α-helices. Beta tubulin is a highly conserved, negatively charged protein, which has made it a popular target for anti-cancer drugs, aimed at disrupting microtubule dynamics and cell division. Anti-beta tubulin antibodies are commonly used as loading controls for immunoblotting to verify that uniform quantities of protein have been loaded across wells.

Figure 3. Immunocytochemistry: HeLa cells were plated at 200 000 cells/ well in 6-well plates on coverslips and allowed to adhere. Following fixation and blocking, cells were incubated with 1:200 dilution of Cape Biologix PtXTM Mouse Anti-β-Tubulin primary antibody (Cat# CBT_A0041), and 1:300 dilution of a commercial Anti-Mouse Alexa Fluor® 488 conjugated commercial secondary antibody. Images were taken on a Zeiss LSM780 with ELYRA PS1 platform confocal microscope (20X) at the Stellenbosch University CAF unit. Thank you to Prof Georgia Schafer (ICGEB) for kindly donating the HeLa cells and Mrs Lize Engelbrecht for her outstanding assistance.
